A thorough examination of existing scholarly works pertaining to the utilization of gene therapy for addressing orthopedic disorders was undertaken. The review underscores the considerable promise of gene therapy in transforming clinical practices within this domain. Orthopedic conditions, encompassing a spectrum of issues such as spinal fusion failure, disc degeneration, fractures, segmental bone defects, bone tumors, articular disorders, soft-tissue injuries, genetic disorders, and nerve and muscular disorders, frequently present challenges for conventional surgical or medical approaches. Consequently, there has been a concerted effort to explore gene therapy as a viable alternative avenue for intervention. Significant progress has been achieved in this innovative approach, although several hurdles must be addressed before its widespread clinical application in humans. Nonetheless, it is evident that gene therapy holds immense promise as a valuable treatment modality for orthopedic disorders in the twenty-first century. As gene transfer techniques continue to advance, the potential applications of gene therapy in orthopedic disorders are expected to expand significantly.
